Indian Biotech Firm Advances Personalised mRNA Vaccines for Cancer Treatment

Can personalised mRNA vaccines offer a new approach to cancer treatment? Hyderabad-based biotechnology firm PopVax is advancing this possibility with the launch of the RNA Foundry, a specialised facility dedicated to developing personalised cancer vaccines using messenger RNA (mRNA) technology.

Backed by the Gates Foundation, Ethereum co-founder Vitalik Buterin, the US government agency BARDA and Facebook co-founder Dustin Moskovitz, the company aims to design and manufacture vaccines tailored to individual cancer patients using genetic information obtained from tumour sequencing.

How Do Personalised mRNA Cancer Vaccines Work?

Unlike conventional cancer treatments, personalised mRNA vaccines target genetic mutations specific to an individual’s tumour. Since these mutations vary from patient to patient, the vaccines use genetic information to train the immune system to recognise and attack cancer cells carrying those mutations.

The technology uses messenger RNA to deliver instructions that help the immune system identify specific cancer targets. This approach builds on the mRNA technology used in several COVID-19 vaccines.

RNA Foundry to Support Vaccine Development

Through its RNA Foundry in Hyderabad, PopVax aims to bring vaccine design and manufacturing together under one roof. The company also plans to leverage the platform and manufacturing processes developed for its broadly protective mRNA COVID-19 vaccine candidate, PVX-001, which has entered clinical trials in Australia.

In addition, its proprietary vaccine-delivery system could offer storage and transportation advantages. According to the company, the system can remain stable for more than nine months under regular refrigeration at 2–8°C, potentially simplifying vaccine distribution.

Clinical Validation Remains a Key Challenge

Although personalised mRNA cancer vaccines have shown promising results internationally, their clinical effectiveness still requires further evaluation. Personalised cancer vaccines developed by US companies Moderna and Merck have demonstrated efficacy against skin cancer in Phase III clinical trials.

However, the effectiveness of vaccines developed through PopVax’s RNA Foundry has yet to be established. The company expects compassionate-use treatments in a small number of patients worldwide to provide early insights and help guide the design of future clinical trials.
